ABSTRACT:

Viruses with capsids larger than 150 nm are considered "giant viruses". Prymnesium kappa virus RFO2 (PkV-RFO2) has a 583 kbp linear dsDNA genome and 200 nm diameter capsid. Using mass spectrometry, we have identified 140 different proteins in purified PkV-RFO2, with nearly 70% of them having unknown function. We used cryo-electron microscopy (cryo-EM) single particle analysis to obtain high-resolution maps of ordered components of the PkV-RFO2 icosahedral capsid, and cross-linking mass spectrometry (XL-MS) to identify protein-protein interactions (PPIs) present in both the icosahedral shell and non-icosahedral core.
